神奇小分子防癌抗衰老 人类距攻克癌症再近一步

美国当地时间5月30日,雅虎、US Press Association、The Morning News等众多美国媒体转载发布霍普金斯大学医学院综合癌症中心肺癌和衰老项目首席科学家张春雨博士最新研究成果《Miraculous Small Molecules - One Step Further towards Helping Cancer Prevention and Healthy Longevity》新闻,译文如下:

  癌症号称死亡之症,人一旦得癌,就像被判了“死缓”,剩下的日子屈指可数。为攻克这一难题,人类一直在进行研究。近日,华裔科学家张春雨博士领导的团队实现重大突破,发现一种新型的转录激活信号通路,可以在癌细胞和正常细胞中,恢复表达兼具抗癌和延缓衰老功效的抑癌蛋白,并将其研究成果转化为膳食补充剂,造福人类。

  世界卫生组织发布的《世界癌症报告》显示,癌症是全球第二大死因,同时40%的癌症是可预防的。“癌症最大的危险因素是年龄,”张春雨指出。

  作为美国霍普金斯大学医学院综合癌症中心肺癌和衰老项目首席科学家,张春雨博士曾在耶鲁、哈佛、国立癌症研究所、拉芙蕾丝呼吸研究所等美国顶级名校和研究机构工作,在“生命时钟”端粒及端粒酶,Werner早衰蛋白与DNA聚合酶,核编码线粒体长寿突变蛋白,老化过程中慢性炎症、肥胖和癌变生物标记物FAT10,免疫调节因子基因表达和组织定位,癌症与衰老关键信号通路PI3K/Akt/mTOR及其抑制剂,肺癌发生过程中基因表达改变与化疗抗性和预后生存,香烟提取物对气道上皮细胞的DNA损伤,NOXA对肺部炎症缓解的分子机制等方面做出了国际领先的研究成果。相关论文在国际顶级期刊及核心专业杂志发表,包括《细胞》、《美国科学院院刊》、《自然》旗下刊物等。

  张春雨博士及其研究团队通过对肿瘤抑制基因RhoB的表达和功能研究,在蛋白水平证明其在维护呼吸系统正常形态和功能中发挥重要作用。他们首次发现,在癌症演化和机体老化过程中,被多种表观遗传学机制下调的肿瘤抑制基因RhoB的高效转录激活的信号通路和分子机制,进而构建了世界上独有的双荧光素单分子载体,并建立了高通量药物筛选系统和创立非侵入肿瘤药物图像实时反应动物模型。此外,该团队还利用该系统从国立癌症研究所多样性小分子库中筛选出新的抗癌小分子,在肺癌、脑瘤和横纹肌肉瘤等多种癌细胞中持续高效诱导肿瘤抑制基因RhoB的蛋白表达,抑制癌细胞增殖、迁移并诱导其凋亡,而没有过多的细胞毒性。

  Dr. Zhang Chunyu and his team studied the expression and function of the tumor suppressor gene RhoB to prove that it plays an important role in maintaining the normal morphology and function of the respiratory system at the protein level. For the first time, they discovered that in the process of cancer evolution and aging, the signaling pathways and molecular mechanisms of efficient transcriptional activation of the tumor suppressor gene RhoB, which are down-regulated by various epigenetic mechanisms, have led to the construction of a unique dual-luciferase single vector in the world. The research team also, establisheda high-throughput drug screening system, and animal models for real-time monitoring anti-tumor drug response. In addition, the team also used the system to screen out several novel anti-cancer small molecules from the National Cancer Institute's Diversity Library, and one of the compounds continuously and efficiently inducesprotein expression of the tumor suppressor gene RhoB in various cancer cells such as lung cancer, brain tumor, and rhabdomyosarcoma,and inhibits cancer cell proliferation, migration and induces apoptosis, without too much cytotoxicity.
